摘要: IL-27是IL-6/IL-12家族细胞因子,可发挥广泛的免疫调节作用.IL-27可通过STAT1依赖或者非依赖途径抑制GATA-3(Th2分化的关键转录因子),从而有利于Th1的分化.IL-27引起naive T细胞产生细胞间黏附分子1,以STAT1依赖方式有利于Th1的分化.IL-27体外还可通过诱导STAT1抑制Th17细胞和iTreg分化,并可诱导T细胞分泌抗炎因子IL-10,减轻病毒免疫应答引起的炎症.在抗病毒反应中,IL-27R信号转导途径对CD8+T细胞T-bet依赖性的IFNγ产生是必不可少的,但急性病毒感染中IL-27R信号系统对CD8+T细胞的增殖来说并不是必需的.近期也有研究认为,IL-27是通过类IFN作用来抑制HCV等病毒复制,但也有人认为,IL-27可能通过IFN依赖和非依赖两种方式抑制HIV的复制.在乙型肝炎患者中,也发现HBV可激活p28和EBI3启动子,并增加IL-27蛋白的产生,但具体临床意义尚待进一步研究.因此,IL-27是一个有潜力的抗病毒因子.此文就IL-27及其受体、IL-27抗病毒的分子机制及其抗病毒作用进行综述. IL-27 belongs to the IL-12 cytokine family,and can widely regulate immune response.IL-27 can suppress GATA-3,a key transcription factor for differentiation of Th2 through STAT1 depended or independed way to facilitate Th1 differentiation.IL-27 can induce intercellular adhesion molecule-1 (ICAM-1 ) and induce Th1 differentiation in STAT1depended way and inhibit Th17 cells and iTreg cells differentiations in vitro.In vivo,IL-27 induce T cells to secrete IL-10,which is an anti-inflammatory agent relieving inflammation in virus immune response.IL-27R signal pathway is essential for IFN γ production by CD8 + T cells in T-bet-depended way in anti-virus response.But in acute viral infection,IL-27R signal pathway is unnecessary,sary for CD8 + T cells proliferation.Recent reserches found that IL-27 can inhibit HCV rephcation via interferon-like function.But others believed IL-27 may inhibit HIV replication though interferon-depended and independed way.In HBV infecters,people found HBV can activate p28 and EBI3 promoters,and induce IL-27 protein.But its clinical meanings need more reseaches.In conclusion,IL-27 is a potential anti-virus factor.IL-27 and its receptor,the molecular mechanisms and anti-virus function of IL-27 are reviewed here.
