目的 明确核小体结合蛋白1(NSBP1)在雄激素非依赖性前列腺癌细胞(PC-3)中的生物学功能.方法 通过RNA干扰技术,使PC-3细胞中的NSBP1表达水平明显降低,然后用胆囊收缩素(CCK)-8法观察细胞活性的变化,用流式细胞光度术检测细胞周期的变化.结果 NSBP 1的表达抑制后,从60 h开始PC-3 细胞活性(A值为0.329±0.017与0±386±0.022)有显著下降(P＜0.05),抑制率到108 h达到高峰(27.5%).流式细胞光度术检测结果显示,NSBP1抑制后的PC-3细胞的G2M+S期细胞百分率也从60 h开始(31.7%±2.0%)与对照组(37.3%±2.5%)有统计学意义(P＜0.05).结论 NSBP 1在前列腺癌PC-3细胞中,对维持细胞活性及促进细胞生长增殖有重要作用.
Objective To investigate biological function of nucleosomal binding protein 1(NSBP1)in androgen-independent prostate cancer cells. Methods Androgen-independent prostate cancer cells of the line PC-3 were cultured and transfected with recombinant inhibiting siRNA expression plasmid pSilencer 2.1-U6 neo.Before transfection and 12,36,60,84,108 and 132 h after transfection RT-PCR and Western blotting was used to detect the inhibition effect. CCK-8 was used to detect the viability of the cells.The cell cycle was examined by flow cytometry. Results The cell viability was significantly reduced since 60-108 h after the transfection(all P＜0.05),the inhibition ratio reaching climax 108 hours after the transfection(P=0.028).However,the inhibition ratio began to increase.FC showed that since 60 h after the transfection the proportion of PC-3 cells at the G2M+S phase began to increased compared to that of the control group ( all P＜0.05).Conclusion NSBP1 has important effect on maintaining cell viability and enhancing growth and proliferation of androgen-independent prostate cancer cells.
