摘要: Homocysteine is one of the major thiols in human plasma and exists in both protein-bound and free thiol forms. It has been shown that homocysteine can modify, through disulfide bonding, class I HLA antigens in the endoplasmic reticulum of the cell. We have also identified CTL, in patients with ankylosing spondylitis or reactive arthritis. that recognised homocysteine-modified HLA B27. A68 and B8. TCR usage by these T cells was analysed by determining the cDNA sequences encoding their TCR alpha- and beta-chains. TCRAV4 and TCRBV23 gene segments were strongly favoured by HLA A68-restricted Hom-CTL from two unrelated patients. The TCR of B37-restricted CTL appeared to be relatively more heterogeneous. although one of the B27-restricted T cell clones employed TCR V gene segments (AV20S1 and BV5S1) very similar to that of the A68-restricted CTL. An arginine or lysine residue at position alpha 93 of the alpha-chain frequently appeared in the TCR of A68- and B8-restricted Hom-CTL, which may suggest a. potential salt-bridge between the carboxyl group of homocysteine and TCR.
