Myrislignan (MRL) existing in nutmeg has been reported to possess inhibition of neoplasm and vascular smooth muscle contraction, and cause a prolonging of hexobarbital-induced hypnosis in mice. In this study, a high-performance liquid chromatography-ultraviolet detection (HPLC-UV) method was developed to analyze MRL in rat tissues. The separation was carried out on an analytical Diamonsil (TM) ODS C-18 HPLC column (250 x 4.6 mm, 5 mu m) equipped with a C-18 guard column (8 x 4 mm, 5 mu m) cartridge system eluted at 1.0 mL min(-1) with a mobile phase methanol-H2O (3:1, v/v). Detection wavelength was 270 nm. The concentration of MRL was analyzed in the brain, testes, muscle, adipose, kidney, heart, lung, stomach, intestine, liver, and spleen at various time points between 8 and 128 min following intravenous administration of MRL to rats at a dose of 20.0 mg kg(-1). The method was successfully used to study the tissue distribution of MRL. The results contribute to explain the effect of nutmeg on the brain, liver and gastrointestinal tract.
