Androgen deprivation therapy, the main method for clinical prostate cancer, was proved to be able to promote the expression of CYP17 alpha 1 gene in adrenal. As CYP17 alpha 1 was the crucial enzyme for the production of 17 alpha-hydroxypregnenolone and dehydroepiandrosterone, a CC-MS method was established to illustrate the change of pregnenolone, 17 alpha-hydroxypregnenolone and dehydroepiandrosterone after orchiectomy in emasculated rat adrenal. Twenty-eight days after orchiectomy, there was a significant increase of 17 alpha-hydroxypregnenolone and dehydroepiandrosterone accompanied with the increase of CYP17 alpha 1 gene expression 14 d after orchiectomy in adrenal.
